Resumen : | The objective of this project was to develop an optimization process using the Response Surface Methodology adjusted with the statistical model of Box Behnken for the extraction of polyphenolic compounds from Opuntia ficus-indica and Opuntia atropes. In addition, the treatments with the highest polyphenolic content and antioxidant capacity were determined, as well as their optimization simultaneously for each species the conditions that favor extraction. The independent variables in this study were: Ethanol (70%, 83% and 96%), Temperature (24 °C, 50 °C and 76 °C) and Time (30 min, 75 min and 120 min). The nopal: solvent ratio was constant at 1:10 p / v. The response variables evaluated were: phenolic acids, flavonoids and antioxidant capacity measured in terms of DPPH• and ABTS•+. For Opuntia ficus-indica, the highest polyphenolic content and antioxidant capacity obtained by simultaneous optimization were the following: phenolic acids 0.921 mg EGA/g, flavonoids 8.799 mg QE/g, DPPH• 0.156 mg EGA/g and ABTS•+ 0.616 mg EGA/g and the process conditions were: ethanol 78.9%, temperature 75.9 °C and time of 120 min. In Opuntia atropes, the highest content of polyphenolic compounds and antioxidant capacity by free radical capture was obtained by simultaneous optimization, and yielded the following yields: phenolic acids 0.978 mg EGA/g, flavonoids 8.368 mg QE/g, DPPH• 0.160 mg EGA/g and ABTS•+ 0.588 mg EGA/g, the independent variables were: 70.0% ethanol, temperature 47.8 °C and time of 120 min.s. El objetivo de este proyecto fue desarrollar un proceso de optimización utilizando la Metodología de Superficie de Respuesta ajustado con el modelo estadístico de Box Behnken para la extracción de compuestos polifenólicos de Opuntia ficus-indica y Opuntia atropes. Además, se determinaron los tratamientos con mayor contenido polifenólico y capacidad antioxidante, así como su optimización de manera simultánea para cada especie las condiciones que favorecen la extracción. Las variables independientes en este estudio fueron: Etanol (70%, 83% y 96%), Temperatura (24 °C, 50 °C y 76 °C) y Tiempo (30 min, 75 min y 120 min). La relación nopal: solvente fue constante a 1:10 p/v. Las variables respuesta evaluadas fueron: ácidos fenólicos, flavonoides y capacidad antioxidante medido en términos de DPPH• y ABTS•+. Para Opuntia ficus-indica el mayor contenido polifenólico y capacidad antioxidante que se obtuvo por optimización simultánea fueron los siguientes: ácidos fenólicos 0.921 mg EAG/g, flavonoides 8.799 mg EQ/g, DPPH• 0.156 mg EAG/g y ABTS•+ 0.616 mg EAG/g y las condiciones de proceso fueron: etanol 78.9%, temperatura 75.9 °C y tiempo de 120 min. |
