Abstract: The excessive use of chemical insecticides for the control of pests has a negative impact on the populations of benefic organisms and can cause the development of resistance. An alternative to this problem is the use of entomopathogens, such is the case of baculoviruses. In the present study, the genetic characteristics and the biological activity of a native isolate of the multiple nucleopolyhedrovirus of Spodoptera frugiperda (SfMNPV) were determined on larvae of its host, S. frugiperda J. E. Smith (Lepidoptera: Noctuidae), the most important maize pest in Mexico. The native virus was genetically compared with another local virus and an isolate from Nicaragua. The genetic comparison was made through the partial sequencing of gene polh. The sequences of the three SfMNPV isolates were aligned and analyzed in a public database using BLAST, which showed a similarity of 99%. To observe the biological activity, bioassays were carried out using third instar S. frugiperda, where a positive relationship between mortality and concentration was observed. The highest mortality was observed at 120 h post-inoculation (hpi) and the adjustment of median lethal concentration (LC50) was obtained at 192 hpi. A production of 1.21 x 10⁸ ± 2.59 x 10⁷ OBs (occlusion bodies)/larva was determined when third instar larvae of S. frugiperda were inoculated with a concentration of 9.5 x 105 OBs/ml. In addition, the effect of a CL40 (4.8 x 104 OBs/ml) on the development and the reproduction of the insect was evaluated.
El uso excesivo de los insecticidas químicos para el control de plagas tiene un impacto negativo en las poblaciones de organismos benéficos y puede causar el desarrollo de resistencia. Una alternativa a esta problemática es el uso de entomopatógenos, tal es el caso de los baculovirus. En el presente estudio se determinaron las características genéticas y la actividad biológica de un aislado nativo del nucleopoliedrovirus múltiple de Spodoptera frugiperda (SfMNPV) sobre larvas de su huésped, S. frugiperda J. E. Smith (Lepidoptera: Noctuidae), la plaga de mayor importancia económica del cultivo del maíz en México. El virus nativo se comparó genéticamente con otro virus local y un aislado procedente de Nicaragua. La comparación se realizó a través de la secuenciación parcial del gen polh. Las secuencias de los tres aislados del SfMNPV se alinearon y se analizaron en una base de datos pública mediante BLAST mostrando una similitud de 99%. Para observar la actividad biológica se realizaron bioensayos con larvas de tercer estadio de S. frugiperda, donde se observó una relación positiva entre la mortalidad y la concentración. La mayor mortalidad se observó a las 120 h post-inoculación (hpi) y el ajuste de la concentración letal cincuenta (CL50) se obtuvo a las 192 hpi. Se determinó una producción de 1.21 x 108 ± 2.59 x 107 OBs (cuerpos de inclusión por sus siglas en inglés)/larva cuando se inocularon larvas de tercer estadio de S. frugiperda con una concentración de 9.5 x 105 OBs/ml. Además, se evaluó el efecto de una CL40 (4.8 x 104 OBs/ml) sobre el desarrollo y reproducción del insecto.
