Zusammenfassung: With increasing antibiotic resistance, there is a continuing need to create new agents to treat infectious diseases of difficult clinical management. In this context, it has developed over 1, 000 antibacterials. Many of them have proven to be toxic in preclinical stages, and just over 100 who are currently on the market are ineffective in at least one of the many antibiotic resistant bacteria. Given this, the medicinal chemistry within the multiple strategies that used, has enabled the development of new organometallic compounds, better known as metaloantibióticos. In this group, the organobismuth complex receives particular attention, for its bacteriostatic and astringent properties. It has also reported organometallic complexes that use as ligands L-α-amino acids, L-α-dipeptides and even peptides with significant antibacterial, antifungal and/or anticancer properties, so therefore represent an active field of research. In this work we synthesized a series of ten dipeptides (L-α-dipeptides) in solution and their corresponding complexes with bismuth salts in a 1:1 ratio, in order to assess their antibacterial activity against strains of clinical interest by the standardized method of disc diffusion (NCCLS). The characterization of complex dipeptides-Bi+3 was performed in solution by nuclear magnetic resonance of carbon and hydrogen (13C and 1H-NMR) in one and two dimensions, and IR spectroscopy. These results suggest that the bismuth in the complex Ala-Phe•Bi+3 interacts with the carboxylate and amide nitrogen forming a ring of five members. In the complex with glycine and alanine in the C-terminal is likely that the bismuth interact with carboxylate, amide nitrogen and the amino N-terminal, forming a bicyclo [3.3.0] pentane.
Con el aumento de la selección de la resistencia antibiótica, surge una continua necesidad de desarrollar nuevos agentes para tratar enfermedades infecciosas de difícil manejo clínico. En este contexto, se han desarrollado más de 1, 000 antibacterianos. Muchos de ellos han demostrado ser tóxicos en fases preclínica, y los poco más de 100 que se encuentran actualmente en el mercado son ineficaces ante al menos una, de las muchas bacterias antibiótico resistentes. Ante esto, la química medicinal, dentro de las múltiples estrategias a las que recurre, ha permitido el desarrollo de nuevos compuestos organometálicos mejor conocidos como metaloantibióticos, en donde los complejos organobismutados reciben particular interés, por sus propiedades bacteriostáticas y astringentes. Así mismo, se han reportado varios complejos organometálicos que utilizan como ligandos L-α-aminoácidos, L-α-dipéptidos e inclusive péptidos, con importantes propiedades antimicrobianas y/o anticancerígenas, por lo que representan un campo activo de investigación. En el presente trabajo se sintetizó una serie de diez dipéptidos (L-α-dipéptidos) en solución y sus correspondientes complejos con sales de bismuto en proporción 1:1, con la finalidad de valorar su actividad antibacteriana frente a cepas de interés clínico por el método estandarizado de difusión en disco (NCCLS). La caracterización de los complejos dipéptidos-Bi+3, se realizó en solución por resonancia magnética nuclear de carbono e hidrógeno (RMN-13C e 1H) en una y dos dimensiones, y por espectroscopía de IR. Dichos resultados sugieren que en el complejo Ala-FenBi+3 el bismuto interacciona con el carboxilato y nitrógeno amida formando un anillo de cinco miembros, y en los complejos con glicina y alanina en el C-terminal es probable que el bismuto además de interactuar con el carboxilato y nitrógeno amida, también interactúe con el amino N-terminal, formando un biciclo [3.3.0] pentano.
