Zusammenfassung: Zooxanthellae are dinoflagellate algae of the genus Symbiodinium which maintain an obligate symbiosis with scleractinian corals. These algae are of the great importance since they provide different nutrients to corals, which allow a higher calcification rate in less time. Currently, nine clades of zooxanthellae have been described, with different physiological and ecological attributes, such as the ability to confer to their hosts some resistance to adverse environmental conditions. To assess the composition and genetic diversity of the zooxanthellae populations present in four coral reefs of the Mexican Pacific after “El Niño” event 2015-2016, zooxanthellae DNA was obtained from 102 fragments of Pocillopora verrucosa coral colonies. DNA was amplified with 28S and ITS2 molecular markers. The resulting phylogenetic trees of both markers showed that 100% of the zooxanthellae’s sequences were grouped with clade D, being closer with subtypes D1.1 and D1.2. The presence of this clade has been related to the ability of host corals to cope with high sea water temperature. Analysis of haplotype networks and genetic diversity showed a greater diversity with ITS2 marker than 28S, with 28 haplotypes vs. 5 haplotypes, respectively. Among the zooxanthellae populations in the four coral reefs, also were found an ancestral haplotype, which haplotypes from Islote Zacatoso moved away with more mutational steps, showing the greatest genetic diversity. These zooxanthellae were harbored in corals that suffered less bleaching during “El Niño” event of 2015 and 2016. In this way, the possible relationship of the current composition of zooxanthellae with their response to the thermal stress caused by this event is discussed: the high genetic diversity of Islote Zacatoso’s zooxanthellae could be related with their high resistance to thermal stress, contrasting with the population of Playa Las Gatas which had the lowest genetic diversity in the area and showed the highest incidence of coral mortality and bleaching.
Las zooxantelas son algas dinoflageladas pertenecientes al género Symbiodinium que mantienen una simbiosis obligatoria con los corales escleractinios. Estas algas son de suma importancia debido a que proveen de diferentes nutrientes a los corales y les permiten desarrollar una mayor tasa de calcificación en un menor tiempo. Existen nueve clados de zooxantelas con diferentes atributos fisiológicos y ecológicos, entre los que están incluidos el conferir a los corales una mayor resistencia a características ambientales adversas. Para evaluar la composición y la diversidad genética de las poblaciones de zooxantelas presentes en cuatro arrecifes coralinos del Pacífico mexicano después del evento “El Niño” 2015-2016, se obtuvo ADN de zooxantelas de 102 fragmentos del coral Pocillopora verrucosa de cuatro arrecifes coralinos de Zihuatanejo, Guerrero. Este material se amplificó y secuenció para los marcadores moleculares 28S e ITS2. Los árboles filogenéticos resultantes de ambos marcadores mostraron que el 100% de las secuencias de Symbiodinium se agruparon con el clado D, siendo más cercanos con los subclados D1.1 y D1.2. A este clado se le ha atribuido resistencia a temperaturas altas. El análisis de redes de haplotipos y de diversidad genética mostraron una mayor diversidad con el marcador ITS2 que con el 28S, revelando 28 haplotipos vs. 5 haplotipos, respectivamente. Entre las zooxantelas de los cuatro arrecifes se encontró un haplotipo ancestral, del que la población de Islote Zacatoso se alejó con más pasos mutacionales, mostrando además la mayor diversidad genética. Los corales con esa población de zooxantelas fueron los que sufrieron menos blanqueamiento durante el evento “El Niño” 2015-16. De esta manera, se discute la posible relación de la composición actual de zooxantelas con un solo clado de zooxantelas es una respuesta al estrés térmico provocado por este evento, pudiendo haber existido anteriormente varios clados de zooxantelas.
