Abstract: This research identified the tolerance to P. cinnamomi of three genotypes of avocado Mexican race (P. americana var. drymifolia), AMUM-064, AMUM-765 and AMUM-773, which were micropropagated from tree explants of the avocado Bank of germplasm of INIFAP-Uruapan (Michoacán, Mexico). Using PCR and qRT-PCR techniques analyzed the presence and expression of candidate genes responsible for tolerance to P. cinnamomi. The micropropagation of the three genotypes was established from axillary buds of adult trees, in the basal MS (half strength) (Murashige and Skoog) with 30 g/L sucrose, 1 g/L activated charcoal, 0.4 mg/L acid ascorbic, 1 g/L commercial fungicide Tecto 60, solidified with 4 g/L of phytagel. Proliferation and elongation of 1-2 shoot/explant to the 45 days of cultivation was achieved in this medium. For the multiplication of shoots, assessed the response in vitro subculturing the shoots in MS with benzyladenine (BA) to 0, 0.25, 0.5, 1.0, 2.5 or 5.0 mg/L in combination with indole-3-butyric acid (IBA) to 0, 0.1, 0.25, 0.5 or 1.0 mg/L. Induction of shoots was observed in a 71.3%, producing 3-5 shoots/explant to the 90 days of the cultivation in MS with 0.5 mg/L of BA and 0.1 mg/L of IBA. For the formation of plantlets, the shoots were rooted in MS with 0.5 mg/L IBA after 90 days of cultivation, the micropropagated plantlets were successfully acclimatized and grown in the greenhouse, keeping them under irrigation every three days and fertilization every three months.
En esta investigación se determinó la tolerancia a P. cinnamomi de tres genotipos de aguacate criollo mexicano (P. americana var. drymifolia), AMUM-064, AMUM- 765 y AMUM-773, los cuales fueron micropropagados a partir de explantes de árboles del Banco de Germoplasma de Aguacate del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias (INIFAP-Uruapan, Michoacán, México). Mediante las técnicas de PCR y qRT-PCR se analizó la presencia y expresión de genes candidatos responsables de la tolerancia a P. cinnamomi. La micropropagación de los tres genotipos se estableció con la siembra de yemas axilares de árboles adultos, en el medio basal MS (Murashige y Skoog) con 30 g/L sacarosa, 1 g/L carbón activado, 0.4 mg/L ácido ascórbico, 1 g/L de fungicida comercial Tecto 60, solidificado con 4 g/L de phytagel. En este medio se logró la proliferación y elongación de 1-2 brotes/explante a los 45 días del cultivo. Para la multiplicación de brotes, se evaluó la respuesta in vitro subcultivando los brotes en MS con benciladenina (BA) a 0, 0.25, 0.5, 1.0, 2.5 ó 5.0 mg/L en combinación con ácido indol-3-butírico (AIB) a 0, 0.1, 0.25, 0.5 ó 1.0 mg/L. La inducción de brotes se observó en un 71.3%, produciendo de 3-5 brotes por explante a los 90 días del cultivo, en MS con 0.5 mg/L de BA y 0.1 mg/L de AIB. Para la formación de plántulas, los brotes fueron enraizados en MS con 0.5 mg/L AIB después de 90 días de cultivo, las plántulas micropropagadas fueron exitosamente aclimatadas y cultivadas en invernadero, manteniéndose bajo irrigación cada tres días y fertilización cada tres meses.
