G-CoA lyase (EC 4.1.3.4) catalyzes the breakdown of HMG-CoA into acetyl- CoA and acetoacetate. In the present work the following parameters of HMG-CoA lyase: has a relative molecular mass of 33 kDa under conditions Denaturants and under native conditions was shown as a homodimer with a Relative molecular weight of 79 kDa. LiuE presented two major isoenzymes with isoelectric points of 6.8 and 7.0, showed a pH and an optimum temperature of 7.0 and 37 ° C, respectively. He showed a non-Michaelian behavior of sigmoidal type, Suggesting an allosteric regulation, the kinetic constants for HMG-CoA were: A K 0.5 740 μM, Vmax of 27.3 U / mg protein and a Hill coefficient of 2.3. Finally it was found that Acetyl-CoA is a competitive inhibitor of HMG-CoA Lyase. The enzymes homologous to LiuE are enzymes specific for their substrate, and Show small affinity constants denoting the high affinity for their substrate, Also possess a Michaelian enzymatic behavior. LiuE has a high HMG-CoA affinity constant relative to its bacterial counterpart (P. Mevalonii) suggesting that it might be able to recognize more than one substrate; these The data reinforced the hypothesis that the enzyme LiuE is a bifunctional enzyme that is Capable of recognizing 3-hydroxy-3-isohexenylglutaryl-CoA (HIHG-CoA) and hydrolyzing it. Using coupled reactions using cis-geranyl-CoA carboxylase (AtuC / AtuF) And isohexenyl glutaconyl-CoA hydrate (AtuE), purified and renatured, were Observed a poor consumption of cis-Geranil-CoA (1.59 nmol consumed) to be absent LiuE in the reaction mixture, whereas when added to the reaction, the Cis-geranyl-CoA increased 11-fold (17.82 nmol cis-geranyl-CoA consumed). With The above evidence is collected to suggest that LiuE has participation in the routes catabolic properties of leucine and acyclic terpenes, by the recognition and Fragmentation of the respective metabolic intermediates (3-hydroxy-3- Methylglutaryl-CoA and 3-hydroxy-3-isohexenylglutaryl-CoA).en
