Resumen : In this work the strain Bacillus sp. UM96 as part of the species B. thuringiensis according to the complete sequencing of the 16S rDNA gene and by the presence of the parasporal body characteristic of this species. The cry gene, which encode the parasporal body, was amplified from the genome of B. thuringiensis UM96. Further, Presented he Gen putative from Endochitinases Chi. The Supernatants of B. thuringiensis UM96 obtained from nutritive broth (CN) And a minimal medium with chitin as the only source of carbon (MQ), showed Chitinase activity, being higher in the MQ supernatant. On the other hand both supernatants showed antifungal activity against Botrytis cinerea, which was similar to that presented by pure commercial chitinase. In addition, both Supernatants incubated at 60°C lost antifungal activity. He MQ supernatant presented higher antifungal activity compared to Supernatant CN. Interestingly, B. thuringiensis UM96 showed Colonization of the rhizosphere in maize plants. Finally, B. thuringiensis UM96 Was able to suppress B. cinerea infection in Medicago truncatula significant way with respect to control plants without any treatment. So, B. thuringiensis UM96 was shown to have a putative gene from the cry1A group, and a chi gene which encodes possibly an endoquitinase, in addition, was found activity of Chitinase activity that could be related in the biocontrol capacity of B. Thuringiensis UM96. With all this, B. thuringiensis UM96 showed high Possibilities to be used as biocontrol of phytopathogenic fungi.
En este trabajo se identificó a la cepa Bacillus sp. UM96 como parte de la especie B. thuringiensis de acuerdo con la secuenciación completa del gen de ADNr 16S y por la presencia del cuerpo paraesporal característico de esta especie. El gen cry, que codifican el cuerpo paraesporal, se amplificó del genoma de B. thuringiensis UM96. Además, presentó el gen putativo de endoquitinasas chi. Los sobrenadantes de B. thuringiensis UM96 obtenidos a partir de caldo nutritivo (CN) y de un medio mínimo con quitina como única fuente de carbono (MQ), mostraron actividad de quitinasa, siendo mayor en el sobrenadante MQ. Por otra parte ambos sobrenadantes mostraron actividad antifúngica contra Botrytis cinérea, la cual fue similar a la que presentó la quitinasa pura comercial. Además, ambos sobrenadantes incubados a 60°C perdieron la actividad antifúngica. El sobrenadante MQ presentó mayor actividad antifúngica en comparación al sobrenadante CN. Interesantemente, B. thuringiensis UM96 mostró capacidad de colonización de la rizósfera en plantas de maíz. Finalmente, B. thuringiensis UM96 logró suprimir la infección de B. cinérea en plantas de Medicago truncatula de manera significativa con respecto a las plantas control sin ningún tratamiento. Así, B. thuringiensis UM96 mostró tener un gen putativo del grupo cry1A, y un gen chi que codifica posiblemente una endoquitinasa, además, se encontró actividad de quitinasa que podría estar relacionada en la capacidad de biocontrol de B. thuringiensis UM96. Con todo esto, B. thuringiensis UM96 mostró altas posibilidades para ser utilizada como biocontroladora de hongos fitopatógenos.
