Resumen : The lactone 6-pentyl-2H-pyran-2-one (6-PP) synthesize is produced by Trichoderma atroviride during its interaction with plants and promotes the emergence of lateral roots and growth of the primary root through auxin-related mechanisms (Garnica-Vergara et al., 2015). MEDIATOR comprises a conserved mutiprotein complex that controls the binding of the enzyme RNA polymerase II and the transcription factors to the gene promoter regions (Carlsten et al., 2013). In this work we found that MEDIATOR 16 is required for the reconfiguration of the root system architecture in response to 6-PP in Arabidopsis. We found that the mutation of the MEDIATOR16 gene (med16-2) increases the length of the primary root and the emergence of lateral roots. Our analyzes revealed that MED16 negatively regulates cell division in the primary root meristem and controls the expression of SHR, a transcription factor involved in the maintenance of the meristem and the regulation of the asymmetric cell division of the cortex/endodermis initial cells. Analysis of the expression of the auxin-related reporter DR5:GFP showed that the negative regulation of MED16 on cell proliferation is independent of auxin accumulation in the root meristem and in lateral root primordia, while the expression lines WOX5:GFP, SHR:GFP and SCR:H2B:YFP in the mutant background med16-2 showed that 6-PP influence the activity of stem cell niche via MED16 in the root meristem.
El compuesto volátil fúngico 6-pentil-2H-piran-2-ona (6-PP) forma parte de las moléculas liberadas por Trichoderma atroviride en su interacción con las plantas y promueve la formación de las raíces laterales y el crecimiento de la raíz primaria a través de mecanismos relacionados con la vía auxínica (Garnica-Vergara et al., 2015). El complejo MEDIADOR es un estructura mutiproteica conservada en los eucariotes que controla la unión entre la RNA polimerasa II y los factores de transcripción a la región promotora de los genes (Carlsten et al., 2013). En este trabajo encontramos que la subunidad MEDIADOR 16 (MED16) es un regulador del efecto de la 6-PP durante la reconfiguración de la arquitectura radicular de Arabidopsis. Los estudios realizados revelaron que MED16 regula negativamente la división celular en el meristemo de la raíz primaria y controla la expresión de SHR, un factor de transcripción involucrado en el mantenimiento del meristemo y la regulación de las divisiones asimétricas de las células iniciales del córtex/endodermis. Los análisis de expresión de la línea reportera de las auxinas DR5:GFP mostraron que la regulación negativa de MED16 sobre la proliferación celular es independiente a la acumulación de auxinas en el meristemo de la raíz primaria y los primordios de las raíces laterales, en tanto que las líneas reporteras WOX5:GFP, SHR:GFP y SCR:H2B:YFP en plantas silvestres y en la mutante med16-2 sugieren que la 6-PP modula la actividad del nicho de células iniciales por medio de MED16 en el meristemo de la raíz primaria.
