Abstract: Background: The obesity is one of the most important problems and occurs due to the excessive accumulation of fat, and it is associated with many diseases such as some types of cancer, type 2 diabetes and cardiovascular disease. The aim of this study was to evaluate various plant extracts as an alternative to prevent and combat obesity for their effect on pancreatic lipase activity. Methods: Plant material was collected from 37 plants to make the extracts, subsequently the degree of inhibition of pancreatic lipase in vitro was compared using orlistat as a control. The plant with the highest inhibitory lipase activity, was evaluate to the estimation of triacylglycerides in plasma of rats, different dose 62.5, 125 and 250 mg/kg of the extract of dry leaves of H. rosea-sinesis was used. Triacyglycerides were measument each hour during three hours. The determination of the lipid lowering effects of H. rosa-sinensis at 125 mg/kg was study in a high-fat diet in Wistar rats, during three weeks. Body weight was measured at weekly intervals. At the end of third weeks the triglycerides and cholesterol levels were also estimated. The acute toxicological test was conducted in Wistar rats. The extract of H. rosa-sinensis was administered in a single dose of 100, 316 and 1000 mg/kg and observed for behavioral changes and mortality. In subacute toxicity studies, Wistar rats were administered the H. rosea-sinensis at dose of 1000 mg/kg via oral dailly for 28 days. Rats were observed if there are any changes in their body weight, food and water intake during this period. Liver and kidney were used to determine biochemical parameters such as GSH, PON 1, MDA and HNE and total proteins.
Antecedentes: La obesidad es uno de los problemas más importantes en la actualidad y se presenta debido a la acumulación excesiva de grasas, se asocia a muchas enfermedades como algunos tipos de cáncer, diabetes tipo 2 y enfermedades cardiovasculares. En el presente trabajo se investigaron varios extractos vegetales como una alternativa para prevenir y combatir a la obesidad mediante la inhibición de la lipasa pancreática. Métodos: Se recolecto material vegetal de 37 plantas para la realización de los extractos, se probó la actividad inhibitoria de la lipasa pancreática in vitro para posteriormente utilizar la planta con mejor inhibición de lipasa en una estimación de la asimilación de triacilglicéridos en animales a diferentes dosis (62.5,125 y 250 mg/kg) midiendo los triacilglicéridos cada hora durante 3 horas, después la estimación de asimilación de triacilglicéridos con una dieta alta en grasa durante tres semanas con la dosis con mejor respuesta en el experimento anterior, se midió el peso semanalmente y al termino se tomó una muestra sanguínea para medir triacilglicéridos y colesterol. El estudio de toxicidad aguda fue con una sola dosis oral a 3 concentraciones (100, 316 y 1000 mg/kg) y se mantuvieron en observación continua durante 72 h y 12 días, por último, la prueba de toxicidad subaguda con la dosis no letal más alta encontrada en el experimento anterior y se administró vía oral diariamente durante 28 días, al final del experimento se extrajo el hígado y los riñones para determinar parámetros bioquímicos como GSH, PON 1, MDA y HNE, además de proteínas totales en cada órgano. Los animales usados en los experimentos fueron Ratus norvergicus cepa Wistar (n=6) (mantenidos en condiciones de bioterio); en los estudios se utilizó el orlistat como estándar positivo de inhibición.
