dc.description.abstractBovine mastitis is the inflammation of the mammary gland and Staphylococcus aureus is the main pathogen responsible, favoring chronic infections. The antibiotic therapy has been deficient as a treatment, showing the need for alternatives such as nisin and chitosan. The objective of this work was to evaluate the immunomodulatory effect of nisin, chitosan, and the nisin-chitosan composite on IIR of bovine mammary epithelial cells (bMECs) infected with S. aureus. The results showed that the growth of S. aureus (ATCC 27543) was not affected by the nisin (1, 2.5, 5 μg/mL; 2 and 24 h), chitosan (10, 25, 50, 75, and 100 μg/mL; 2 and 24 h), or the nisin/chitosan composite (1/10, 2.5/10 and 5/10 μg/mL; 2 and 24 h). Only nisin at concentrations >10 μg/mL (24 h) affected the growth of the bacteria. The nisin (1, 2.5, 5, and 10 μg/mL; 24 h), chitosan (10, 25, 50, 75, and 100 μg/mL; 24 h) or the composite (1/10, 2.5/10, and 5 /10 μg/mL; 24 and 48 h) did not affected the viability of the bMECs. Also, the nisin, chitosan, or composite did not modify the nitric oxide (NO) production of the bMECs. Nisin (1 μg/mL) together with the infection, significantly increased the production of NO. RT-qPCR analysis showed that the chitosan (10 μg/mL) induced IL-1β expression (~2-fold). On the other hand, the nisin (1 μg/mL), chitosan (10 μg/mL), or the composite (1/10 μg/mL), together with the infection, induced the expression of IL-10 (~2, ~4, and ~5 -fold, respectively). Likewise, they reversed the repression of IL-8 exerted by the infection. In addition, the composite favors a higher expression of DEFB1 (~3-fold) that the exerted by the separate molecules. In conclusion, the nisin, chitosan, or the composite differentially modulate the expression of IIR-related genes in bMECs infected with S. aureus.en
dc.description.abstractLa mastitis bovina es la inflamación de la glándula mamaria y Staphylococcus aureus es el principal patógeno responsable, favoreciendo infecciones crónicas. La terapia con antibióticos ha sido deficiente como tratamiento, lo que muestra la necesidad de alternativas, como la nisina y quitosano. El objetivo del presente proyecto fue evaluar el efecto inmunomodulador de nisina, quitosano y el compósito nisina-quitosano sobre la RII de células epiteliales mamarias bovinas (CEMB) infectadas con S. aureus. Los resultados mostraron que el crecimiento de S. aureus (ATCC 27543) no se afectó por la nisina (1, 2.5, 5 μg/mL; 2 y 24 h), quitosano (10, 25, 50, 75 y 100 μg/mL; 2 y 24 h) o el compósito nisina-quitosano (1/10, 2.5/10 y 5/10 μg/mL; 2 y 24 h). Solo la nisina a concentraciones >10 μg/mL (24 h) afectó el crecimiento de la bacteria. La nisina (1 μg/mL) durante la infección, aumentó significativamente la producción de NO. Los resultados de RT-qPCR mostraron que el quitosano (10 μg/mL) indujo la expresión de la IL-1β (~2 veces). Por otro lado, la nisina (1 μg/mL), quitosano (10 μg/mL) o el compósito (1/10 μg/mL), durante la infección, incrementaron la expresión de la IL-10 en ~2, ~4 y ~5 veces, respectivamente. Así mismo, estos compuestos revirtieron la represión sobre la expresión de la IL-8 ejercida por la infección. Además, el compósito favoreció la expresión de la DEFB1 (~3 veces), ya que fue superior a la ejercida por las moléculas por separado.es_MX
