Título : Microbiota intestinal de juveniles y adultos Chirostoma estor (Piscis: Atherinopsidae) en ambiente silvestre y en cultivo

Resumen : The microbiota "dominated by bacteria" fulfills important functions in fish such as the stimulation of the immune system and the synthesis of metabolites related to growth. In general, the microbiota associated with organisms is influenced by changing factors such as the habitat and the stage of development of the host, and in particular, the intestinal microbiota of fish in controlled culture can be altered by the application of prebiotic and probiotic supplements to the diet. These supplements are also involved in improving growth and survival, thanks to the fact that they activate the immune system. In this study, the intestinal microbiota of the whitefish Chirostoma estor in the adult state from wild environments (Lake Pátzcuaro) and controlled environment (culture) is characterized for the first time, thus determining the baseline of the bacterial microbiota, which later allows to study its dynamics in nutritional or environmental experiments. Also presented in this work is a nutritional experiment carried out for 12 weeks where prebiotics (inulin from Agave tequilana, cell wall of Saccharomyces cerevisiae) and synbiotics (from the English synbiotics, meaning pre- and probiotics simultaneously) were applied: Lactobacillus acidophilus + inulin and L. acidophilus + cell wall, to the diet of juvenile C. estor to determine their effects on the intestinal microbiota and growth. These microbiomic studies were carried out by metagenomics of amplicons, extracting DNA from digesta and anterior and posterior intestine of adult wild and farmed fish (N=74), and intestine of juvenile fish fed with prebiotics and synbiotics (N=42), to immediately perform Illumina Miniseq sequencing of the V3 region of the 16S rRNA gene and bioinformatic analysis of the generated amplicons.
La microbiota "dominada por bacterias" cumple funciones importantes en peces como es la estimulación del sistema inmune y la síntesis de metabolitos relacionados con crecimiento. En general, la microbiota asociada a organismos es influenciada por factores cambiantes como son el hábitat y la etapa de desarrollo del hospedero, y en particular, la microbiota intestinal de peces en cultivo controlado puede ser alterada por la aplicación de suplementos prebióticos y probióticos a la dieta. Estos suplementos también intervienen en la mejora del crecimiento y supervivencia, gracias a que activan el sistema inmune. En este estudio se caracteriza por primera vez la microbiota intestinal del pez blanco Chirostoma estor en estado adulto proveniente de ambientes silvestre (Lago de Pátzcuaro) y ambiente controlado (cultivo), determinando así la línea base de la microbiota bacteriana, lo que posteriormente permite estudiar su dinámica en experimentos nutricionales o ambientales. También se presenta en este trabajo, un experimento nutricional llevado a cabo durante 12 semanas en donde se aplicaron prebióticos (inulina de Agave tequilana, pared celular de Saccharomyces cerevisiae) y simbióticos (del inglés synbiotics, significando pre- y probióticos simultáneamente): Lactobacillus acidophilus + inulina y L. acidophilus + pared celular, a la dieta de juveniles de C. estor para determinar sus efectos en la microbiota intestinal y en el crecimiento. Estos estudios microbiómicos se realizaron por metagenómica de amplicones, extrayendo ADN de digesta e intestino anterior y posterior de peces adultos silvestres y cultivados (N=74), e intestino de peces juveniles alimentados con prebióticos y sinbióticos (N=42), para enseguida realizar secuenciación por Illumina Miniseq de la región V3 del gen ARNr 16S y realizar análisis bioinformático de los amplicones generados.
