The cryopreservation of post-mortem sperm seems to be an effective tool to conserve the genetic material of endangered species, native or native species, as well as animals of high zootechnical value, that die unexpectedly. The aim of the present work is to cryopreserve bovine spermatozoa obtained from the tail of the epididymis (CE) and vas deferens (CD) of post-mortem bulls. A total of 10 intact testes were collected from animals of municipal trail. The sperm were obtained from the EC and CD using the retrograde flow technique. The recovered sperm were kept in a commercial TRIS-based diluent. Progressive Mobility (MP), Viability (V) and Abnormal Morphology (MA) were evaluated. Subsequently they were frozen by the conventional method, using a concentration of 50 x 106 sperm per dose. The doses were thawed at 37ºC in a water bath for one minute to consecutively evaluate the MP and V. The preliminary results were analyzed using a generalized linear model considering the treatments as a fixed effect, the differences between means were evaluated using the technique of minimum differences significant for fresh CE and CD sperm, the MP average was 71.7 ± 2.6 and 75.0 ± 4.5; the V was 68.7 ± 4.84 and 79.2 ± 5.6; the MA was 57.27 ± 5.9 and 42.33 ± 6.02, respectively. For EC sperm and post-thaw CD, the MP observed was 41.0 ± 7.04 and 55.7 ± 8.71 respectively. These data indicate that there are no significant differences between the sperm extracted from the EC and CD, with respect to the MP and V post-thawing rate.
La criopreservación de espermatozoides obtenidos post-mortem parece ser una herramienta eficaz para conservar el material genético de especies en peligro de extinción, especies autóctonas o nativas, así como de animales de alto valor zootécnico, que mueran inesperadamente. El objetivo del presente trabajo es criopreservar espermatozoides de bovino obtenidos de la cola del epidídimo (CE) y conducto deferente (CD) de toros post-mortem. Se colectaron un total de 10 testículos intactos provenientes de animales de rastro municipal. Los espermatozoides se obtuvieron de la CE y CD utilizando la técnica de flujo retrógrado. Los espermatozoides recuperados se mantuvieron en un diluyente comercial a base de TRIS. Se evaluó la Movilidad Progresiva (MP), Viabilidad (V) y Morfología Anormal (MA). Posteriormente se congelaron por el método convencional, utilizando una concentración de 50 x 106 de espermatozoides por dosis. Las dosis se descongelaron a 37ºC en baño María durante un minuto para consecutivamente evaluar la MP y V. Los resultados preliminares se analizaron empleando un modelo lineal generalizado considerando a los tratamientos como efecto fijo, las diferencias entre medias se evaluaron empleando la técnica de diferencias mínimas significativas. Para los espermatozoides de la CE y CD frescos el promedio de MP fue de 71.7 ± 2.6 y 75.0 ± 4.5; la V fue de 68.7 ± 4.84 y 79.2 ± 5.6; la MA fue de 57.27 ± 5.9 y 42.33 ± 6.02, respectivamente. Para los espermatozoides de la CE y CD post-descongelación, la MP observada fue de 41.0 ± 7.04 y 55.7 ± 8.71 respectivamente. Estos datos nos indican que no existen diferencias significativas entre los espermatozoides extraídos de la CE y CD, respecto a la tasa de MP y V post-descongelación.
