In the present investigation, the objective was to identify the fungus that causes the death of avocado trees with symptoms of white rot. Several avocado orchards in Michoacan, Mexico, with symptoms of the disease, were sampled; basidiomas of Hydnopolyporus spp. were observed, associated with the root system and the trunk of the trees. Samples of basidiomas from different municipalities and areas were taken and evaluated for their characters: morphological, molecular, and pathogenic (symptom severity and virulence). The morphological and taxonomic identification was made from the basidiomas collected in the field, taking into account the macromorphological data of size, consistency, color, all in the different parts of the basidioma (cap, context, hymenophore and stipe) as well as their cultural characteristics in PDA culture medium. In the molecular identification, the primers ITS (ITS4/ITS5), LSU (LOR0f/LR5r), RPB2 (Brpb2-6r2/Brpb2-7r2) and EF-1α (EF1-BF1/ EF1-B-R) were used for the tests. For pathogenicity, healthy avocado seedlings of Persea americana Mill. Var. Drymifolia. From the samples taken in the field, 19 pure strains identified morphologically as Hydnopolyporus fimbriatus were obtained. These colonies developed a cottony and corky type of growth, in some cases prostrate to the culture medium. Subsequently, the pathogenicity of the strains was evaluated, the first symptoms in the avocado seedlings appeared 14 days after their inoculation, all the isolates produced symptoms like those observed in the field, the H5 and H8 strains caused the death of the seedlings. At 77 days. Four gene fragments were obtained for each sample and were used for molecular identification and in the analysis, by Blast comparison the species Hydnopolyporus fimbriatus was identified with 99% identity. Phylogenetically, with all the concatenated gene fragments, it was grouped into a single monophyletic clade, related to the genera Emmia, Flavodon and Irpex.
En la presente investigación el objetivo fue identificar al hongo causante de la muerte de árboles de aguacate con síntomas de pudrición blanca. Se muestrearon varios huertos de aguacate en Michoacán, México, con síntomas de la enfermedad, se observaron basidiomas de Hydnopolyporus spp., asociados al sistema radicular y el tronco de los árboles. Se tomaron muestras de basidiomas de diferentes municipios y zonas los cuales fueron evaluados por sus caracteres: morfológicos, moleculares y de patogenicidad (severidad y virulencia de los síntomas). La identificación morfológica y taxonómica se realizó a partir de los basidiomas recolectados en campo, se tomaron en cuenta los datos macromorfológicos de tamaño, consistencia, color, todo ello en las diferentes partes del basidioma (píleo, contexto, himenoforo y estípite) así como sus características culturales en medio de cultivo PDA. En la identificación molecular se usaron los primers ITS (ITS4/ITS5) LSU (LOR0f/LR5r), RPB2 (Brpb2-6r2/Brpb2-7r2) y EF-1α (EF1-B-F1/EF1-B-R), para las pruebas de patogenicidad se utilizaron plántulas de aguacate sanas de Persea americana Mill. var. drymifolia. De las muestras tomadas en campo se obtuvieron 19 cepas puras identificadas morfológicamente como Hydnopolyporus fimbriatus, estas colonias desarrollaron un crecimiento tipo algodonoso y corchoso, en algunos casos postrado al medio de cultivo. Posteriormente se evaluó la patogenicidad de las cepas los primeros síntomas en las plántulas de aguacate se presentaron a los 14 días a partir de su inoculación, todos los aislamientos produjeron síntomas como los observados en campo, las cepas H5 y H8 causaron la muerte de las plántulas a los 77 días. Se obtuvieron cuatro fragmentos de genes por cada muestra y fueron utilizados para la identificación molecular y los análisis, por comparación Blast se identificó la especie Hydnopolyporus fimbriatus. Filogenéticamente con todos los fragmentos de genes concatenados se agrupo en un solo clado monofiletico, emparentado con los géneros Emmia, Flavodon e Irpex.
