In traditional medicine it is recognized that the Neem tree (Azadirachta indica), has numerous healing properties for various ailments. With respect to the pharmacological actions reported it has been shown to have antioxidant, antitumor, anticancer, pro-apoptotic, antiproliferative, antibacterial, caries, antifungal properties. In the search for active ingredients have been found to exist oid, however, they have not been reported as active principles proteins. To our knowledge, there are no reports of lectins on this floor. For this reason, and represent a source of therapeutic products aimed at this work, I was raised to study the presence of lectins in leaf extracts of Neem, as these are involved in many physiological and pathological processes both. activity was analyzed using hemagglutination assay and identifying lectins started in the neem tree. The crude extract present hemagglutination activity for the four human blood groups: A1 and A2: GalNAc-Gal-Fuc, B: Gal-Gal-Fuc and O: Fuc-Gal. It is indicating that the extract contains a mixture of lectins. Once this presence of lectins was tested. It began with an enriched mixture using a higher initial protein concentration and soluble pigments and eliminating compounds, which was called protein fraction and the initial results obtained with the crude extract was confirmed. Subsequently, to identify the affinities and proportions of this mixture of lectins, we proceeded to perform a fractional precipitation using ammonium sulfate intervals from 0 to 100%, within the results it shows that in the range 0-20% has an affinity preferred for blood group O, at intervals of 20-40%, 40-60 have preferential affinity for A2.
En la medicina tradicional se reconoce que el árbol del Neem (Azadirachta indica), posee numerosas propiedades curativas para diversos padecimientos. Con respecto a las acciones farmacológicas reportadas se ha comprobado que tiene propiedades antioxidantes, antitumorales, anti cancerígenas, pro-apoptosis, antiproliferativo celular, antibacterial, caries, antifúngica. En la búsqueda de principios activos se ha encontrado que existen liminoides, sin embargo, no se han reportado como principios activos a las proteínas. Hasta donde sabemos, no existen reportes de lectinas en esta planta. Por esta razón, y por representar una fuente de productos terapéuticos, como objetivo de este trabajo, se planteo estudiar la presencia de lectinas en extractos de hojas del Neem, ya que estas intervienen en numerosos procesos tanto fisiológicos como patológicos. Se analizó la actividad utilizando un ensayo de hemaglutinación y se inició la identificación de las lectinas presentes en el árbol del Neem. El extracto crudo presento actividad hemaglutinante para los cuatro grupos sanguíneos humanos: A1 y A2: GalNAc-Gal-Fuc, B: Gal-Gal-Fuc y O: Fuc-Gal. Lo que indica que el extracto contiene una mezcla de lectinas. Una vez que se comprobó esta presencia de lectinas. Se partió de una mezcla enriquecida, utilizando una mayor concentración inicial de proteínas y eliminando los pigmentos y compuestos liposolubles, a la que se le llamó fracción proteica y en la que se corroboró los resultados iniciales obtenidos con el extracto crudo. Posteriormente, para identificar las afinidades y proporciones de esta mezcla de lectinas, se procedió a realizar una precipitación fraccionada utilizando sulfato de amonio con intervalos de 0 a 100%, dentro de los resultados se observa que en el intervalo 0-20% tiene una afinidad preferente para el grupo sanguíneo O, en los intervalos de 20-40%, 40-60 tienen afinidad preferente con A2.
