Purificación y Caracterización de lectinas de capitaneja Verbesina croata

Abstract

The capitaneja (Verbesina crocata) is used in traditional Mexican medicine against gynecological conditions, digestive disorders, diarrhea, typhoid, dysentery, cancerous tumors, intestinal fevers, among others. Recent research has shown its hypoglycemic activity. As far as is known, there are no reports of lectins in this plant, so this research work represents the first study of these proteins in it. For this, the hemagglutinating activity of its crude extract, protein fraction and 5 different ranges of salt concentration in the ABO System in Humans, as well as certain blood groups of animals, were determined. The crude extract obtained specific activity in the four blood groups, mainly: an activity of 237.449 ± 4.424 UHE/mg with blood group B, with the blood of animals: the highest hemagglutinating activity was observed with rabbits, Spanish horses, frisol and quarter mile with 234,057 UHE/mg. Of the different fractions analyzed, the 20-40% and 60-80% fractions presented the highest protein quantity as well as the highest hemagglutinating activity for group B: 230,400 ± 22,897 UHE/mgL(20-40%) and 240,941 ± 0UHE/ mgL (60-80%). These fractions were subjected to size exclusion chromatography and electrophoresis.

La capitaneja (Verbesina crocata)es utilizada en la medicina tradicional de México contra afecciones ginecológicas, trastornos digestivos, diarrea, tifoidea, disentería, tumores cancerígenos, fiebres intestinales, entre otras. Investigaciones recientes han demostrado su actividad hipoglicémica. Hasta donde se sabe no existen reportes de lectinas en esta planta por lo que el presente trabajo de investigación representa el primer estudio de estas proteínas en ella. Para ello se determinó la actividad hemaglutinante de su extracto crudo, fracción proteica y 5diferentes intervalos de concentración salina en el Sistema ABO en Humanos, así como determinados grupos sanguíneos de animales. El extracto crudo obtuvo actividad específica en los cuatro grupos sanguíneos, mayoritariamente: una actividad de 237,449 ± 4,424 UHE/mg con el grupo sanguíneo B, con la sangre de animales: la mayor actividad hemaglutinante se observó con el cunícola, equino español, frisol y cuarto de milla con 234,057 UHE/mg. De las diferentes fracciones analizadas las fracciones 20-40% y 60-80% presentaron la mayor cantidad proteica así como la mayor actividad hemaglutinante para el grupo B de: 230,400 ± 22,897UHE/mgL(20-40%) y 240,941 ± 0UHE/mgL (60-80%). Estas fracciones fueron sometidas a cromatografías de exclusión molecular y electroforesis.
