Diabetes is a metabolic disease often associated with high blood pressure. Currently these diseases are a pandemic and the leading cause of death in the world. Several drugs are used to treat it, but all have side effects and none are totally effective. Lipoic acid (LA) has been shown in several studies to have hypoglycemic, antioxidant, and anticancer effects. LA is essential for cells, being a cofactor of various enzymes involved in aerobic metabolism. Therefore, in the present work we investigated the effect of hypoglycemic, hypolipidemic and on vascular reactivity of LA in diabetic rats. Diabetes was induced with a single dose of streptozotocin intraperitoneally (IP) in male Wistar rats (220g + 20g). There were 4 groups of 6 animals each: control group (1), control group + LA (35 mg/kg), administered via IP for 15 days (2), diabetic group (3), diabetic group + LA (4). At the end of the treatment, glucose tolerance and insulin resistance curves were performed and triacylglycerides (TG) were quantified. Arterial contraction in isolated aortas in response to phenylephrine. We observed a hypoglycemic effect comparing group 4 against group 3, as well as a decrease in TG. A vasorelaxant effect was also observed in the aortas of group 4 respect to group 3. Therefore, by presenting a hypoglycaemic and hypolipidemic effect, LA can help the treatment of DM and by its vasorelaxant effect it can also be useful for treatment of arterial hypertension.
La diabetes es una enfermedad metabólica frecuentemente asociada a la hipertensión arterial. Actualmente estas enfermedades son una pandemia y la principal causa de muerte en el mundo. Para su tratamiento se usan diversos fármacos, pero todos producen efectos secundarios y ninguno es totalmente efectivo. Se ha demostrado en varios estudios que el ácido lipoico (AL) tiene efectos hipoglucemiantes, antioxidantes y anticancerígenos. El AL es esencial para las células, al ser cofactor de diversas enzimas involucradas en el metabolismo aeróbico. Por lo que en el presente trabajo investigamos el efecto hipoglucémico, hipolipemiante y sobre la reactividad vascular del AL en ratas diabéticas. La diabetes se indujo con una dosis única de estreptozotocina vía IP en ratas Wistar macho (220g ±20g). Se tuvieron 4 grupos con 6 animales cada uno: grupo control (1), grupo control + AL (35 mg/kg), administrado vía IP durante 15 días (2), grupo diabético (3), grupo diabético + AL (4). Al terminar el tratamiento se realizaron curvas de tolerancia a la glucosa, de resistencia a insulina y se cuantificaron los triacilglicéridos (TG). También se determinó, la contracción arterial en aortas aisladas en respuesta a la fenilefrina. Observamos un efecto hipoglucémico comparando el grupo 4 contra el grupo 3, así como una disminución en los TG. También se observó un efecto vasorrelajante en las aortas del grupo 4 con respecto al 3. Por lo tanto, al presentar un efecto hipoglucémico e hipolipemiante, el AL podría ayudar al tratamiento de la DM y por su efecto vasorrelajante también podría ser útil para el tratamiento de la hipertensión arterial.
