Colorectal cancer is an important public health issue because is one of the most frequent cancers and have a high mortality rate. The search of bioactive compounds from plants with anticancer and chemopreventive properties have a prominent place in research against this disease. Callistemon citrinus is used as ornamental in Mexico. Recent studies have shown that has a wide range of biological properties with possible therapeutic applications, highlighting the antioxidant activity. The main objective of this study was to determine the protective effect of ethanolic extracts of C. citrinus of leaves and flowers in a model of colorrectal cancer induced in rats. The extracts were analyzed phytochemically and was identified 20 chemical compounds, mainly composed of terpenic. Eucalyptol was the most abundant compound in leaves (7.5%) and 3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one (DMPP) in flowers with 4.4%. This compound had not previously been reported in the literature for C. citrinus. Both extracts presented higher content of total phenolic and flavonoids compounds. The antioxidant capacity in vitro of the extracts was evaluated by three assays: ABTS, DPPH and FRAP. The extracts of flowers presented a higher antioxidant capacity than the leaves extracts. The extracts were tested in a model of colorectal cancer induced with 1,2 dimethylhydrazine (DMH) in rats. Four experimental groups (n = 8) were established: one control, and three that were administered with 5 doses (65 mg/kg) of DMH.
El cáncer colorrectal es uno de los tipos de cáncer más frecuentes y con mayor tasa de mortalidad por lo que se ha convertido en un grave problema de salud pública. La búsqueda de compuestos bioactivos con propiedades anticancerígenas y quimiopreventivas ocupa un lugar destacado en la investigación contra este padecimiento. Callistemon citrinus es una planta utilizada en México como ornamental. Estudios recientes han demostrado que posee una amplia gama de propiedades biológicas con posibles aplicaciones terapéuticas, destacando la actividad antioxidante. El objetivo principal de este estudio fue determinar el efecto protector de extractos etanólicos de hojas y flores de C. citrinus en un modelo de cáncer de colon inducido en ratas. Los extractos fueron analizados fitoquímicamente y se identificó la presencia de 20 compuestos químicos, la mayoría de tipo terpénico. El eucaliptol fue el compuesto más abundante en hojas (7.5%) y la 3-dihidro-3,5-dihidroxi-6-metil-4H-piran-4-ona (DMPP) en flores con 4.4%. Este compuesto no había sido reportado previamente en la literatura para C. citrinus. Ambos extractos presentaron un alto contenido de fenoles totales y flavonoides. La capacidad antioxidante in vitro de los extractos fue evaluada por tres técnicas: ABTS, DPPH y FRAP. Los extractos de flores presentaron una mayor capacidad antioxidante que los extractos de hojas. Los extractos fueron probados en un modelo de cáncer colorrectal inducido con 1,2 dimetilhidrazina (DMH) en ratas. Se establecieron cuatro grupos experimentales (n=8): uno control, y tres que fueron administrados con 5 dosis de 65 mg/kg de DMH.
