Polyphenolic compounds have been used as antioxidants and food pigments, as well as food supplements due to their outstanding antioxidant capacity and their organoleptic properties. Most of these compounds are obtained from plant species cultivated in a conventional manner, where their production is subject to the species and phenological stage of the plant, as well as to various biotic and abiotic factors that limit their production. Therefore, techniques such as in vitro culture have been used to control the physicochemical conditions of incubation and development, making it an effective technique for the controlled synthesis of phenolic compounds and their optimization. In this context, the objective of the present work was to evaluate the response in the production of polyphenols, flavonoids and antioxidant capacity under different stimuli in callus coming from the leaf and stem of Rubus glaucus. For this, 18 treatments were designed based on MS medium with 1 mg. L-1 2,4-D combined with different concentrations of sucrose (15, 30 and 60 g • L-1), proportion of potassium nitrate (0, 1, 2) and mineral salts (50 and 100%); 7 g • L-1 of agar and 1 g • L-1 of PVP. The experimental design was completely randomized with a factorial arrangement of 2 x 3 x 3. An analysis of variance was made with multiple comparison of means with a confidence interval of 95%.
Los compuestos polifenólicos han sido utilizados como antioxidantes y pigmentos alimentarios, así como suplementos alimenticios esto debido a su destacada capacidad antioxidante y sus propiedades organolépticas. La mayoría de estos compuestos son obtenidos de especies vegetales cultivadas de forma convencional, donde su producción se ve sujeta a la especie y etapa fenológica de la planta, así como a diversos factores bióticos y abióticos que limitan su producción. Por lo que, se ha recurrido a técnicas como el cultivo in vitro, que permite controlar las condiciones fisicoquímicas de incubación y desarrollo, convirtiéndola en una técnica eficaz para la síntesis controlada de compuestos fenólicos y su optimización. En este contexto, el objetivo del presente trabajo fue evaluar la respuesta en la producción de polifenoles, flavonoides y capacidad antioxidantes bajo diferentes estímulos en callo proveniente de hoja y tallo de Rubus glaucus. Para ello, se diseñaron 18 tratamientos a base de medio MS con 1 mg. L-1 de 2,4- D combinado con diferentes concentraciones de sacarosa (15, 30 y 60 g•L-1), proporción de nitrato de potasio (0, 1, 2) y sales minerales (50 y 100 %); 7 g•L-1 de agar y 1 g•L-1 de PVP. El diseño experimental fue completamente al azar con arreglo factorial de 2 x 3 x 3. Se realizó un Análisis de varianza con comparación múltiple de medias con un intervalo de confianza del 95 %.
