Adrenergic receptors (AR) belong to the family of heterotrimeric G proteincoupled receptors. AR α1B, a plasma membrane protein that participates in vasoconstriction and cell proliferation, may function as a proto-oncogene when its coupling to G protein is altered. When RA α1B is stimulated with a catecholamine, such as epinephrine, the expression of several cell cycle regulatory genes is altered; specifically, there is an increase in the expression of genes involved in the G1/S transition of the cell cycle, causing its deregulation. Studies show the eukaryotic translation initiation factor 3, subunit f (eIF3f) physically interacts with AR α1B. The synthesis of eIF3f is also related to the cell cycle, its expression is circumscribed to specific moments of the cell cycle, such as the G2/M transition, the S phase, and in a lesser extent, the early G1 phase. However, the function of the eIF3f-RA α1B interaction and its involvement in the control of cell division are unknown. Therefore, it is interesting to study the interaction dynamics of these two proteins during the different phases of the cell cycle. Objective: To determine the expression profile of AR α1B and its interaction with eIF3f during the cell cycle of human A549 cells.
Los receptores adrenérgicos (RA) pertenecen a la familia de receptores acoplados a proteínas G heterotriméricas. El RA α1B, una proteína de membrana plasmática que participa en la vasoconstricción y proliferación celular, puede funcionar como un protooncogen cuando se altera su acoplamiento con la proteína G. Cuando se estimula al RA α1B con una catecolamina, como epinefrina, se altera la expresión de varios genes reguladores del ciclo celular; específicamente, se da un aumento en la expresión de genes involucrados en la transición G1/S del ciclo celular, provocando la desregulación del mismo. Estudios demuestran que la subunidad f del factor eucariótico de iniciación 3 (eIF3f) interactúa físicamente con el RA α1B. La síntesis de eIF3f también está relacionada con el ciclo celular, está circunscrita a puntos específicos como a la transición G2/M, a la fase S y, en menor medida, a la fase G1 temprana. Sin embargo, se desconoce la función de la interacción eIF3f-RA α1B y si está involucrada en el control de la división celular, por lo que resulta interesante estudiar la dinámica de interacción de estas dos proteínas durante las distintas fases del ciclo celular. Objetivo: Determinar el perfil de expresión del RA α1B y la interacción de éste con eIF3f durante el ciclo celular de células humanas A549.
