The maize, Zea mayz L., is one of the most culturally and economically important crops in Mexico. This crop is mainly affected by the fall armyworm, Spodoptera frugiperda (J. E. Smith). Environmentally compatible alternatives are required for its control. Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV) (Baculoviridae) is a specific virus capable of generating high mortality rates towards its host. The present study evaluated the in vivo production of a native isolate of SfMNPV (YUC-SfMNPV) on fourth instar S. frugiperda fed with two types of diets: a standard agar diet (D1) and another prepared with fiber soybean (D2) (Objective 1). The post-mortem larval weights (68.5 ± 1.70 and 62.8 ± 1.71 mg) and the production of the infective particles (occlusion bodies, OBs) per larva (4.15x107 ± 1.91x106 and 5.24x107 ± 7.03x106) were not significantly affected by the D1 and D2, respectively. In addition, an experiment was performed to evaluate the optimal time of virus harvest (OBs per mg of biomass) using D1 (Objective 2). The number of OBs per mg of larvae was statistically equal between the 2 and 3 days and between 5- and 6-days post-inoculation, highest OB production at the last two days was observed (3.80 and 2.78 x105 OBs/mg larva, respectively). On the other hand, experiments were carried out to quantify and identify total fatty acids (FAs) in healthy and infected S. frugiperda larvae with the YUC-SfMNPV isolate at two post-inoculation times (24 and 144 h) (Objective 3).
El maíz, Zea mayz L., es uno de los cultivos con mayor importancia cultural y económica en México. Este cultivo se ve principalmente afectado por el gusano cogollero, Spodoptera frugiperda (J. E. Smith). Para su control se requieren alternativas compatibles con el ambiente. El nucleopoliedrovirus múltiple de Spodoptera frugiperda (SfMNPV) (Baculoviridae) es un virus específico capaz de generar mortalidades elevadas hacia su huésped. El presente estudio evaluó la producción in vivo de un aislamiento nativo del SfMNPV (YUC-SfMNPV) sobre larvas de cuarto estadio de S. frugiperda alimentadas con dos tipos de dietas: una estándar elaborada con agar (D1) y otra elaborada con fibra de soya (D2) (Objetivo 1). Los pesos de las larvas post-mortem (68.5 ± 1.70 y 62.8 ± 1.71 mg) y la producción de partículas infectivas (cuerpos de inclusión, CI) por larva (4.15x107 ± 1.91x106 y 5.24x107 ± 7.03x106) no fueron significativamente afectados por las D1 y D2, respectivamente. Además, se realizó un experimento para evaluar el tiempo óptimo de cosecha del virus (CI por mg de biomasa) utilizando la D1 (Objetivo 2). El número de CI por mg larva fueron estadísticamente iguales entre los días 2 y 3 y entre los días 5 y 6 post-inoculación, siendo estos dos últimos días los de mayor producción (3.80 y 2.78 x105 CI/mg larva, respectivamente). Por otro lado, se realizaron experimentos para cuantificar e identificar los ácidos grasos (AGs) totales en larvas de S. frugiperda sanas e infectadas con el aislamiento YUC-SfMNPV en dos tiempos post-inoculación (24 y 144 h) (Objetivo 3).
